【摘要】Objective:ToexploretherelationshipofCDH17expressionwith

clinico-pathologicalfeaturesandthecorrelationbe-tweenthesingle

nucleotidepolymorphisms(SNPs)ofCDH17geneandgenetic

susceptibilityofgastriccarcinoma(GC).Methods:Atissuemicroarraywas

performedtosimulatethedynamicprocessofinvasionandmetastasisof

GC.Immunohistochemicalstainingwasperformedtodetectthe

expressionofCDH17protein,andPCR-basedLDRwasperformedto

detectthe2SNPloci(rs2514813andrs3214050)genotypesofCDH17

gene.Results:TheexpressionofCDH17proteininGCwasmore

significantlyup-regulatedandgreatlyincreasedintheintestinaltypethan

inthediffusetype.TheexpressionofCDH17proteininGCwaspositively

correlatedwiththehistologicalgrading(P0.01)andwasnotassociated

withthesurvival(P=0.209).Withtheprogressionofthecancerinvasion,

theexpressionofCDH17proteininGCshowedadowntrendfromthe

gastricmucosalayertotheinvasivefrontedge.ThefrequenciesoftheC

andTallelesandtheCC,CT,andTTgenotypesattheCDH17rs3214050

locusbetweentheGCpatientsandthecontrolgroupsweresignificantly

different(P0.01).However,nosignificantdifferenceswereobservedat

rs2514813(P0.05).TheindividualswiththeTal-leleshadlongersurvival

timethanthosewiththeCCgenotype(P0.01).Conclusion:Theup-

regulationofCDH17expressionisin-volvedinthemaintenanceof

histologicalphenotypeandprogressionofGC.IndividualswithTallelesat

theCDH17rs3214050locushavedecreasedriskofGCandhadbetter

prognosis(OR=0.762,95%CI:0.619-0.937),therebysuggestingthat

screeningforthesealleleswouldhelpwiththeassessmentofgenetic

susceptibilityandprognosisofGCintheFujianpopulation.%目的:探讨
