幽门螺杆菌IgG抗体胶体金免疫层析快速诊断试纸条的研制 【摘要】? 目的? 建立一种快速检测抗H.pylori IgG抗体的胶体金免疫层析试纸条，并优化制备中各关键步骤的实验条件。方法? 以柠檬酸三钠还原法制备20nm的胶体金溶液，葡萄球菌A蛋白（SPA），制备免疫胶体金复合物，组装胶体金免疫层析快速诊断试纸条。结果? 用柠檬酸还原法制备的20nm胶体金溶液呈亮红色。胶体金标记SPA的最低稳定浓度5μg/ml，最适稳定量为6μg/ ml。SPA标记的最适pH为6.0。以此试纸条检测 HYPERLINK /class_free/121_1.shtml 临床血清标本与进口ELISA试剂盒比较，组间阳性率比较差异无显著性。结论? 胶体金免疫层析试纸条质量稳定性不但与抗原抗体的选择、用量优化、层析 HYPERLINK /class_free/150_1.shtml 材料的选择、胶体金的制备与标记等因素密切相关，而且缓冲系统的优化、辅助添加剂的选择与优化组合也非常重要。 ??? 【关键词】? 幽门螺杆菌；胶体金；免疫层析检测?? 　　Research on development of gold immunochromatographic test kit for serum H.pylori antibody IgG 　　【Abstract】? Objective? Developing gold immunochromatographic test kit for serum H.pylori antibody IgG and optimize the key experiment conditions.Methods? Preparing the colloid gold particles of 20 nm which were coupled with Streptococcus protein A (SPA)，set up the gold immunochromatography test kit for serum H.pylori antibody IgG.Results? The spheroidal colloid gold particles of 20nm is shown in color of bright red.The minimal stable concentration (MSC) of gold- SPA is 5μg/ml，and the suitable stable concentration ( SSC) is 6μg/ml.The pH 6.0 is appropriate. 53 sera samples were tested by GICA and ELISA，there is on significant difference between two methods.Conclusion? The quality of gold immunochromatography test kit is associated with the factors，such as the quality and quantity of antigen or antibody， colloid gold particles，buffers，etc. 　　【Key words】? helicobacter pylori;colloid gold;gold immunochromatographic assay ??? 在幽门螺杆菌（Helicobacter pylori，Hp）感染的诊断中，细菌培养、组织病 HYPERLINK /class_free/12_1.shtml 理学检查、尿素酶试验、呼气试验等方法或因为给患者造成的侵入性痛苦，或需要一定的条件和技术，或因为需要特殊仪器，或因为费用昂贵等，其临床推广及多次重复追踪复查受到限制。而血清学检测因其非侵入性、快速、准确，可同时处理大量标本，检测不同类型的抗体，在临床 HYPERLINK /class_free/121_1.shtml 检验、基础研究中倍受青睐。20世纪80年代兴起的胶体金免疫层析检测（gold immunochromagraphy assay，GICA），操作简单快捷、结果清晰易于判断、无需复杂的实验技能和特殊设备，特别适于基层卫生单位使用。本研究预采用H.pylori超声全菌体抗原、葡萄球菌A蛋白（SPA）抗原，尝试研制一种根据双抗原夹心法检测原理检测血清、唾液中抗H.pylori的IgG抗体的胶体金免疫层析检测试剂，优化胶体金免疫层析快速诊断试纸条研制各关键步骤的实验条件，为进一步研制和开发奠定实验基础。 　　1? 材
