烟草土壤微生物PLFAs特征及其多样性分析 张秋芳1,唐莉娜2,史 怀1,杨述省1,周先冶1,林营志1,刘 波1* (1福建省农科院农业生物资源研究所,福建 福州 350003; 2福建省烟草公司,福建 福州350003) 摘要:磷脂脂肪酸(PLFAs)法是一种可用于定性和定量分析土壤微生物群落多样性的方法。本研究应用PLFAs法分析烟草土壤微生物磷脂脂肪酸特征,并结合Shannon-Wiener(H1)、Brillouin(H2)、 Mcintosh多样性指数(H3)、丰富度指数(S)和Pielou均匀度(J)、Simpson优势度指数(D)等多样性指数测度方法,对烟草土壤微生物群落多样性、丰富度和均匀度进行分析研究。初步研究结果表明,不同生长阶段供试烤烟土壤微生物群落中,先后出现的PFLAs有43种,其多样性指数、丰富度和均匀度皆不相同;聚类分析结果表明,可把Shannon-Wienner指数与土壤微生物PFLAs含量分类相似的结果相结合,将烟草土壤中的PFLAs分成四大类:Ⅰ类为高含量高多样性,Ⅱ类高含量中多样性(过渡型),Ⅲ类中含量高多样性(过渡型),Ⅳ类低含量低多样性。 关键词:烟草土壤;微生物群落;磷脂脂肪酸(PLFAs);多样性 The characteristics of phospholipid fatty acids(PLFAs)and its diversity of microbial community in the tobacco soils (ZHANG Qiu-fang 1, TANG Li-na 2, SHI Huai1, Yang Shu-sheng1, ZHOU Xian-zi1, Lin Ying-zhi1 and LIU Bo 1* (1 Agricultural Bioresource Institute,Fujian Academy of Agricultural Sciences,Fuzhou,Fujian 350003,China ; 2 Fujian Tobacco Company,Fuzhou,Fujian 350003 China) Abstract:Phospholipid fatty acids(PLFAs)is one of the methods to analysis the microbial community diversity qualitatively and quantitatively. The characteristics of phospholipid fatty acids of tobacco soils was analyst, combining with Shannon-Wiener(H1),Brillouin(H2) and Mcintosh(H3) diversity index,the abundance(S),Pielou evenness(J)and Simpson index(D).The total 43 kinds of PFLAs in soils which were tested in different growing periods of tobacco were all not similar at the microbial community diversity, the abundance and evenness.But there was some similarity at the result of cluster analysis between Shannon-Wiener(H1)diversity index and the relevant content of PLFAs,and then the 43 kinds of PFLAs were divided to 4 classifications as follows:NO.1 High quantity and high diversity;NO.2 High quantity and moderate diversity(transition type); NO.3 Moderate quantity and high diversity(transition type); NO.4 Low quantity and low diversity。 Key words:Tobacco soil; Microbial community; Phospholipid fatty acids(PLFAs); Diversity 前言 土壤微生物以其丰富的生物多样性使它们成为生态系统中最活跃和最具影响力的组分之一,土壤微生物多样
