Methods the cortical neurons of SD fetal rats were cultured for 10~12 days, respectively, with glutamate and mechanical scarification. Immunohistochemical double labeling reaction was performed by SP. Results 2 hours after glutamate, the c-fos protein began to express and was obviously concentrated in the nucleus after 4 hours. 2 hours after trauma only in the wound edge direct injury c-fos protein expression, away from the wound without direct injury of neurons appeared in 4 hours after trauma; a few neurites were distal c-fos protein particles; 6 hours after injury of hippocampal neurons began to disintegrate, but c-fos protein particles had not disappeared. Conclusion glutamate injury can be induced by the expression of c-fos protein in cultured neurons, injury of cultured cortical neurons and glial cells can release neurotoxic substances, cultured neurons after injury c-fos expression may be associated with apoptosis.
