水迷宫训练对抑郁大鼠齿状回神经干细胞增殖和
血管内皮生长因子表达的影响

摘要 目的 研究抑郁症学习记忆障碍与齿状回神经干细胞增殖和血管内皮生长因子(VEGF )表达的关系。方法 20 只雌性
Wistar 大鼠随机分为对照组和抑郁症模型组(模型组)。复制慢性不可预见性应激模型。旷场实验评价大鼠行为学变化,Morris
水迷宫定位航行实验评估大鼠学习记忆能力,电镜观察大鼠海马结构及其微小血管形态学变化,免疫组化技术检测Nestin 及
VEGF 表达。结果 与对照组比较,模型组大鼠的行为学水平和垂直活动得分显著降低;定位航行实验逃避潜伏时和游泳距离
显著延长;海马微小血管口径变大,管壁变厚,内皮细胞结构不规则;Nestin 免疫阳性细胞数和VEGF 表达均明显低于对照组。
结论 抑郁症学习记忆障碍与慢性应激导致的海马微小血管损伤、海马齿状回神经再生抑制有关。

Vascular Endothelial Growth Factor in Depression Model Rats

Abstract Objective To study the relationship between learning and memory disorder of depression and neural⁃stem⁃cell proliferation and expres⁃
sion of vascular endothelial growth factor (VEGF ). Methods A total of 20 female Wistar rats were randomly divided into control group and depres⁃
sion model group (model group ). A chronic unpredicted stress model was established. The open field method was used to assess behavioral scores of
rats. The Morris water maze navigation experiment was adopted to evaluate the learning and memory ability of rats. An electron microscope was used
to observe the morphological change of hippocampal structure and its vessels. Immunohistochemistry was used to detect expressions of Nestin and
VEGF in hippocampal neurons. Results Compared with those in control ,in the model group ,horizontal and vertical scores were reduced markedly ,
