18β甘草次酸衍生物对smmc-7721的影响 Effects of 18β-glycyrrhetinic Acid Derivatives in SMMC-7721 cells 摘要:目的:研究18β-甘草次酸衍生物对SMMC-7721、细胞增殖的影响。方法:利用细胞计数法,倒置显微镜法,MTT法,oecHhst-33258染色法,流式细胞仪检测法,观察和研究经不同浓度18β-甘草次酸衍物作用24h,48h和72h的肝癌细胞SMMC-7721的形态结构,生长抑制率,细胞凋亡的变化,并探究了对SMMC-7721影响的最适18β-甘草次酸衍物的浓度。结论:经不同浓度的18β-甘草次酸衍物作用后的SMMC-7721,经显微镜观察显示:细胞的形态发生改变,出现膨胀或皱缩现象;通过MTT检测表明细胞的增殖抑制率随18β-甘草次酸衍物浓度的增加而增加,随着作用时间的增长而增加,通过,Hoechst-33258试剂盒染色法研究表明18β-甘草次酸衍物还可影响细胞的凋亡。 关键词:18β-甘草次酸衍物SMMC-7721 细胞增殖 细胞凋亡 细胞形态 Effects of 18β-glycyrrhetinic Acid Derivatives in SMMC-7721 cells Abstract: Purpose: Study the 18β-glycyrrhetinic Acid Derivatives to cancer of the liver the influence of cells. Methods: Using the method of microscopic to inverted microscope, MTT method, Hoechst-33258 Staining Kit. In order to observations and research the 18β-glycyrrhetinic Acid Derivatives training 24 h, 48 h and 72h of liver cancer cells SMMC-7721 the morphological structure, growth inhibition rate and the change of the cell apoptosis. And we will explore the effect of SMMC-7721 on the most suitable 18β-glycyrrhetinic acid derivative of concentration.
