大花君子兰总生物碱抗肿瘤研究于妮娜包海鹰吉林农业大学中药材学院,长春130118 摘要:采用MTT法观察大花君子兰总生物碱(简写为TACMR)对体外培养的肺癌细胞NCI-H446、胃癌细胞SGC-7901、乳腺癌细胞MCF-7和肝癌细胞SMMC-7721的增殖抑制作用用小鼠移植性肿瘤法检测TACMR对小鼠H22肝癌的作用,并通过脾指数和胸腺指数等指标综合考察TACMR对肿瘤小鼠免疫机能的影响。结果:TACMR对NCI-H446、SGC-7901、MCF-7和SMMC-7721均具有明显的细胞增殖抑制作用,且均呈现良好的量效关系TACMR作用于4种不同肿瘤细胞48 h的IC50值分别为38.55,12.08,8.85,7.04 μgmL;体内试验结果表明,TACMR对H22肝癌具有一定的抑制作用。TACMR剂量在34 mgkg时,抑率为35.2%脾指数与相比显著(P<0.01)。TACMR剂量在136 mgkg时,抑率仅为3.7%,胸腺指数和脾指数明显减少(P<0.01),与胸腺指数(P<0.01)。TACMR具有作用,同时对免疫功能有一定的影响。关键词:大花君子兰;总生物碱;MTTH22荷瘤小鼠 (9.5仿宋) 中图分类号(小五黑体): A   文章编号(小五黑体):1000-5684 2011 DOI(小五黑体): 网络出版地址(小五黑体): Anti-Tumor Effect of Total Alkaloids from Clivia miniata Regel (小三号Times New Roman 加粗) YU Ni-na , BAO Hai-ying,HE Zhong-mei(11号Microsoft JhengHei) College of Chinese Medicinal Materials, Jilin Agricultural University,Changchun 130118,China(5号Times New Roman) Abstract(5号Arial): In vitro, the inhibitory effects of total alkaloids from Clivia miniata Regel TACMR on proliferation of NCI-H446, SGC-7901, MCF-7, and SMMC-7721 were measured by MTT colorimetric assay. The inhibitory effects of TACMR on H22 hepatic cancer was observed in mice with subcutaneous inoculation on the tumor cells and its effect on immunological function of mice bearing tumor by observing the influence to spleen index, thymus index and so on. In vitro, the results of MTT showed that TACMR inhibited proliferation of NCI-H446, SGC-7901, MCF-7, SMMC-7721 and a good dose-effect relationship and the IC50 values on them were 38.55, 12.08, 8.85 and 7.04μg/mL. In vivo, TACMR has significant inhibitory effect on H22 tumor cells. The inhibition ratio was 35.2% with the dosage of 34 mg/kg, compared with the positive control group, spleen index increased significantly P<0.01 .
