摘 要 从明永冰川低温土壤中分离尧纯化获得一株低温假单胞菌噬菌体PFV1 并对其生物学特性(噬菌体基

Isolation and Characterization of a Lytic Cold-active
Bacteriophage of Pseudomonas f luorescens

Abstract :A lytic cold-active bacteriophage, designated as PFV 1, was isolated from soil sample of Mingyong
glacier.Restriction fragment length polymorphism analysis of genomic DNA, constituent proteins and physio原
logical analysis of PFV 1 were further carried out to characterize the phage. Based on the 16S rRNA gene se原
quence, the host strain was identified as a member of Pseudomonas f luorescens . The morphology of the iso原
lated phage was observed by electron microscope. The bacteriophage is spherical, about 50 nm in diameter.
Physiological analysis results showed that PFV 1 had infection activity at 4 , and plaques were formed be原
tween 4 and 25 . It was sensitive to chloroform, and showed thermolability. Furthermore, the genome of
PFV 1 consists of double-stranded DNA and is approximately 38 kb.
