融合中的作用

【摘要】 目的评价慢病毒转染的骨髓间充质干细胞诱导大鼠脊柱融合的能力。方法构建携
带骨形态发生蛋白2(BMP.2)的慢病毒并将其转染到骨髓间充质干细胞中,观察细胞的碱性磷酸酶染
色和活性。并将转染的细胞移植到大鼠脊柱融合模型中,8周后处死大鼠,对其脊柱标本进行手触力
学测试,Micro—CT扫描和组织学切片观察。结果慢病毒转染细胞的碱性磷酸酶染色明显好于未转染
细胞(3.2±0.4比1.1±0.2,P0.05)。转染的细胞移植到大鼠体内8周后,x线、Micro.CT、手触力学测试
和组织学切片均显示转染的大鼠脊柱标本全部达到骨性融合。而未转染的大鼠脊柱标本均未融合。
结论慢病毒转染的骨髓间充质干细胞能够诱导脊柱融合,可作为基因载体工具用于脊柱融合。
