ISG15蛋白稳定表达细胞系的构建及其与猪源BVDV--2相互作用初步研究的开题报告

【摘要】

ISG15蛋白是一种干扰素-stimulated基因(ISG)产物，可以调节免疫反应和细胞增殖，并在许多病毒感染和细胞外压力中发挥重要作用。本研究旨在构建ISG15稳定表达细胞系，并初步研究其与猪源BVDV-2的相互作用。

通过经经过筛选后的侧翼肌细胞(ISG15+/+)，构建ISG15稳定表达细胞系，并采用Westernblot鉴定ISG15的表达水平。同时，利用ELISA和Real-timePCR进行猪源BVDV-2的检测和分析，探究ISG15蛋白与猪源BVDV-2的相互作用特点。

通过本研究的开展可为进一步探索ISG15蛋白的功能及其在病毒感染发生过程中的分子机制提供新思路和参考。

【Abstract】

ISG15proteinisaproductofinterferon-stimulatedgenes(ISGs),whichcanregulateimmuneresponsesandcellproliferation,andplayimportantrolesinmanyvirusinfectionsandextracellularstresses.TheaimofthisstudyistoconstructanISG15stableexpressioncelllineandtopreliminarilystudyitsinteractionswithporcineBVDV-2.

ISG15stableexpressioncelllineswereconstructedbysidemusclecells(ISG15+/+)thathadbeenscreened.TheexpressionlevelofISG15wasidentifiedbyWesternblot.Meanwhile,ELISAandReal-timePCRwereusedfordetectionandanalysisofporcineBVDV-2toexplorethecharacteristicsofinteractionsbetweenISG15proteinandporcineBVDV-2.

Bycarryingoutthisstudy,newideasandreferencesareprovidedforfurtherexplorationofthefunctionofISG15proteinanditsmolecularmechanismintheprocessofvirusinfection.
