江医学2013 年第35 卷第17 期 miR-142-3p 在结直肠癌中的 表达及其临床意义 余红敏 积贤 任约翰 薛迪新 林道浙 余铭 林肖 渠宁 【摘要 】 目的 探讨结直肠癌组织中miR-142-3p 的表达及其与临床病理特征的关系。 方法 分别提取42 例结直肠癌 患者癌组织及癌旁组织标本中的总RNA ，以RT-PCR 及荧光定量 PCR 技术测定miR-142-3p 的表达量，并分析miR-142-3p 的 表达与肿瘤的侵袭、转移 临床病理特征及预后的关系。结果 结直肠癌组织中miR-142-3p 的表达低于对应的癌旁组织[-2. 652(-0.469~-4.420) vs -1.046 (-0.488~-2.589)] ，癌旁组织中miR-142-3p 的表达是癌组织中的3.258 倍，两者具有统计学差 异（P＜0.05 ），miR-142-3p 的低表达与结直肠癌淋巴结转移、临床分期有关(均P＜0.05) ；而与患者的年龄、性别、肿瘤大小、远处转 移、肿瘤位置、肿瘤浸润深度、分化程度无关（均P ＞0.05 ）。结论 miR-142-3p 在结直肠癌的发生、发展中可能起着抑癌基因的 作用，有望成为结直肠癌诊断与治疗的另一新指标。 【关键词】 结直肠癌 miR-142-3p 荧光定量PCR 抑癌基因 Expression of miR-142-3p in human colorectal cancer and its clinical significance China 【Abstract 】 Objective To investigate the expression of miR-142-3p in colorectal cancer and its clinicopathological sig- nificance. Methods Surgical specimens of cancer tissues and adjacent normal mucosa were obtained from 42 patients with colorectal cancer. Total RNA of cancer tissues and adjacent normal mucosa were extracted, and the relative expression levels of miR-142-3p were detected by quantitative real-time reverse transcriptase polymerase chain reaction (RT-PCR). The correla- tion of miR-142-3p with tumor invasion, metastasis and other clinicalpathologic parameters was analyzed. Results The rela- tive expression levels of miR-142-3p were significantly lower in colorectal cancer tissues than those in normal mucosa [-2.652 (-0.469~-4.420) vs -1.046 (-0.488~-2.589), P ＜0.05]. The expression of miR-142-3p was negative correlated with the lymph node metastases and clinical stage (P ＜0.05); there was no significant correlation of miR-142-3p with age, gender, tumor size, site, distant metastases, depth of tumor invasion, differentiation degree (P ＞0.05). Conclusion The expression of miR-142-3p may play a tumor suppressor role in the development of colorectal cancer, it
