Introduction:
Nasopharyngealcarcinoma(NPC)isahighlyinvasiveandmetastaticmalignancythatoccurspredominantlyinSouthernChinaandSoutheastAsia.Despitetheadvancementsintreatments,theprognosisofpatientswithadvancedormetastaticNPCisstillpoor.Therefore,itiscrucialtoidentifynoveltargetsandpathwaysthatmayserveaspotentialtherapeuticinterventionsforNPC.Theextracellularsignal-regulatedkinase/mitogen-activatedproteinkinase(ERK/MAPK)signalingpathwayisacrucialregulatorofcellgrowth,proliferation,andsurvival,whichhasbeenlinkedtovarioushumancancers.However,theexactroleoftheERK/MAPKsignalingpathwayintheproliferationofNPCcellsremainsunclear.
Objective:
ThisstudyaimstoinvestigatetheroleoftheERK/MAPKsignalingpathwayintheproliferationofNPCcellsandexploreitspotentialasatherapeutictargetforNPC.
Methods:
1.Cellcultureandtreatment:NPCcelllineswillbeculturedandtreatedwiththemitogen-activatedproteinkinase/extracellularsignal-regulatedkinase(MEK)inhibitor,U0126,toinhibittheERK/MAPKpathwayactivation.
2.Cellviabilityassay:CellCountingKit-8(CCK-8)assaywillbeusedtomeasurecellviabilityandproliferation.
3.Westernblotanalysis:Theproteinexpressionsofp-ERKandERKwillbedetectedbyWesternblotting.
4.Statisticalanalysis:Theresultswillbestatisticallyanalyzedusingone-wayANOVAfollowedbyDunnett’smultiplecomparisontest.
ExpectedResults:
WeexpectthattheinhibitionoftheERK/MAPKpathwaybyU0126willreducetheproliferationofNPCcells.Furthermore,weanticipateasignificantdecreaseintheexpressionofp-ERKintheU0126-treatedcellscomparedtothecontrolgroup.
