PAGE 8 题目:双乙酰反应在联二酮   测定中的应用 题 目:双乙酰反应在联二酮测定中的应用 题目:双乙酰反应在联二酮测定中的应用 摘 要 双乙酰是常温下浅黄色液体,容易挥发,稀溶液有奶油气味,主要使用于调制奶油和香精。双乙酰是一种联二酮。啤酒,酸奶等大众饮品的风味主要受双乙酰的影响。如何科学的简便的测定双乙酰含量是一个值得研究的问题。我们利用原理,当1-萘酚存在时,含胍基的化合物与双乙酰(联二酮)反应,在强碱性介质中,生成红色化合物,在一定浓度范围内红色化合物的含量与其浓度符合朗伯-比尔定理,故可应用此原理测定双氰胺(DCD)或联二酮的含量。本文我们主要研究联二酮,本次研究分别应用2,3-丁二酮、2,3-戊二酮、2,3-己二酮和2,3-庚二酮为测定目标,探讨了双乙酰分光光度法测定联二酮含量的最佳显色时间及碱液、催化剂、显色剂的最佳用量,确定了不同显色体系的线性范围、工作曲线和检出限。 本文的研究结果表明: 1)2,3-丁二酮最佳显色条件:加样体积1ml,比色波长540nm,加碱体积2ml,催化剂体积5ml,显色剂体积3ml。 2)2,3-戊二酮最佳显色条件:加样体积2ml,比色波长544nm,加碱体积2ml,催化剂体积5ml,显色剂体积3ml。 3)2,3-己二酮最佳显色条件:加样体积2ml,比色波长536nm,加碱体积2ml,催化剂体积5ml,显色剂体积3ml。 4)2,3-庚二酮最佳显色条件:加样体积2ml,比色波长546nm,加碱体积2ml,催化剂体积6ml,显色剂体积3ml。 实验结果还表明4种显色体系的回收率均在97.7 %~101.0%之间,RSD在1.97 %~6.57 %之间,方法具有较高的准确度和精密度。 关键词:双乙酰,双乙酰反应,分光光度法 The application of the reaction for diacetyl in the determination of VDK ABSTRACT Diacetyl is pale yellow liquid at room temperature, volatile, with butter smell when the state is dilute solution, mainly used for modulation of cream and spices. Diacetyl is a kind of VDK. The flavor of beer, yogurt and other popular drinks is mainly affected by diacetyl. How to determination diacetyl scientific and handy is a problem worthy of study. We use the principle of when catalyst 1-naphthol exist that the reaction between arc group-containing compounds with diacetyl in strong alkaline medium form a red compound. Within a certain range of concentrations with a linear relationship between absorbance. Accordingly established for the determination of diacetyl content spectrophotometry. We used 2,3-butanedione, 2,3-pentanedione, 2,3-hexanedione and 2,3-hetanedione as chromogenic agent to determine VDK, discussed the time of color reaction and the optimal dose of alkali, catalyst and chromogenic agent, and determined linearity range, calibration curve and detection limit under different colorimetric methods.
