苦参碱抑制人雄激素非依赖性PC3细胞增殖和诱导凋亡的研究

【摘要】 目的 研究苦参碱对人雄激素非依赖性前列腺癌细胞株PC3增殖、细胞周期及细胞凋亡的影响。方法 不同浓度苦参碱作用PC3细胞后,应用MTT法观察苦参碱对PC3细胞增殖的影响,流式细胞仪研究苦参碱对PC3细胞周期的影响,TUNEL法、Annexin V/PI双染分析苦参碱对PC3细胞凋亡的影响。结果 苦参碱对PC3细胞的生长具有抑制作用,与对照组相比有统计学差异(Plt;0.05)。苦参碱对PC3细胞的增殖抑制作用呈现时间及剂量依赖性关系。苦参碱的最佳药物浓度为1.0g/L,最佳作用时间为48h。流式细胞仪分析显示不同浓度苦参碱均可使PC3细胞阻滞于G0/G1期,随着苦参碱浓度的增加,G0/G1期细胞所占的比例增加,与对照组相比有统计学差异(Plt;0.01)。TUNEL法、Annexin V/PI分析显示不同浓度苦参碱均可诱导PC3细胞凋亡,其早期凋亡率及晚期凋亡率随着苦参碱浓度的增加而升高,呈现剂量依赖性关系(Plt;0.05,Plt;0.01)。结论 苦参碱在体外可抑制PC3细胞的增殖,具有时间和浓度依赖关系。抑制增殖和细胞周期阻滞与G0/G1期相关。苦参碱可诱导PC3细胞凋亡,与苦参碱的作用浓度及时间有关。

ABSTRACT: Objective To investigate the effects of matrine on the proliferation, cell cycle, apoptosis of androgen independent prostate cancer (AIPC) cell line PC3 in vitro. Methods PC3 cells were treated by different concentrations of matrine. The inhibitory effect of matrine on the proliferation of PC3 cells was observed by MTT method. The cell cycle of PC3 cells treated by matrine was observed by flow cytometric assay. Apoptosis of PC3 cells induced by matrine was determined with TUNEL and Annexin V/PI staining assay. Results MTT test showed that PC3 cells treated by matrine had an inhibitory effect, which was significantly different from that of controls (Plt;0.05). Matrine markedly inhibited cell proliferation of PC3 in dose and timedependent manners. The optimal dose of matrine which inhibited PC3 cells was 1.0g/L and the best time of inhibition was at 48h. Flow cytometric assay showed that the PC3 cells treated by different doses of matrine were arrested in cell cycle progression at G0/G1 phase. The cell ratio at G0/G1 phase increased with the increase of matrine dose, which was significantly different from that of controls (Plt;0.01). TUNEL and Annexin V/PI staining assay showed that matrine could obviously induce the apoptosis of PC3 cells.
