Background:Glycogensynthasekinase-3β(GSK-3β)isaserine/threonineproteinkinasethatisinvolvedinawiderangeofcellularprocesses.Itisakeyenzymeinglycogensynthasemetabolism,insulinsignaling,andcellcycleregulation.DysregulationofGSK-3βhasbeenimplicatedinseveraldiseases,includingtype2diabetes,Alzheimersdisease,andcancer.Therefore,understandingthestructureandfunctionofGSK-3βisofsignificantimportanceindevelopingtargetedtherapiesforthesediseases.
Objective:TheaimofthisstudyistoinvestigatethedynamicpropertiesoftheGSK-3βproteinusingmoleculardynamicssimulations.Specifically,wewillfocusonunderstandingthestructuralchangesandfunctionalimplicationsofGSK-3βphosphorylationbyexaminingitsinteractionswithsubstrates,inhibitors,andotherregulatoryproteins.
Methods:WewillperformmoleculardynamicssimulationsonthecrystalstructuresofGSK-3βinitsactiveandinactivestates.Thesimulationswillbecarriedoutusingdifferentforcefieldsandconditionstoexplorethepotentialenergylandscapeandconformationalspaceoftheprotein.
