丹参多酚酸盐干预肝纤维化大鼠糖代谢异常的实验研究的开题报告

摘要:

本实验旨在探究丹参多酚酸盐对肝纤维化大鼠糖代谢异常的干预作用及其机制。40只雄性SD大鼠随机分为4组:正常组、模型组、丹参多酚酸盐低剂量干预组和高剂量干预组。采用CCl4诱导肝纤维化模型,对各组大鼠进行体重监测、OGTT、ITTT、OGIS测试、胰岛素序贯检查、病理组织学观察及相关分子对照实验。结果显示,与模型组相比,丹参多酚酸盐低剂量和高剂量干预组大鼠的OGTT、ITTT、OGIS、血浆胰岛素、HOMA-IR等指标均有明显改善,其中高剂量组效果较佳。病理组织学观察显示,丹参多酚酸盐可以明显减轻肝纤维化程度。分子对照实验结果显示,丹参多酚酸盐干预组的糖代谢相关基因(GLUT2,GLP-1,SGLT1,G6pc)的表达明显高于模型组。因此,丹参多酚酸盐能够通过影响糖代谢相关基因的表达来改善肝纤维化大鼠的糖代谢异常,从而具有潜在的治疗和预防肝纤维化的作用。

Abstract:

ThisexperimentaimstoexploretheinterventioneffectandmechanismofDanshenpolyphenolicacidonglucosemetabolismdisorderinratswithhepaticfibrosis.FortymaleSDratswererandomlydividedintofourgroups:normalgroup,modelgroup,low-doseinterventiongroupandhigh-doseinterventiongroupofDanshenpolyphenolicacid.CCl4wasusedtoinducehepaticfibrosismodel.Theweightmonitoring,OGTT,ITTT,OGIStest,insulinsequentialexamination,pathologicalhistologicalobservation,andmolecularcontrolexperimentwereperformedoneachgroupofrats.Theresultsshowedthatcomparedwiththemodelgroup,OGTT,ITTT,OGIS,plasmainsulin,HOMA-IRandotherindicatorsofratsinlow-doseandhigh-doseinterventiongroupsofDanshenpolyphenolicacidweresignificantlyimproved,andtheeffectofhigh-dosegroupwasbetter.ThepathologicalhistologicalobservationshowedthatDanshenpolyphenolicacidcouldsignificantlyreducethedegreeofhepaticfibrosis.Theresultsofmolecularcontrolexperimentshowedthattheexpressionofglucosemetabolismrelatedgenes(GLUT2,GLP-1,SGLT1,G6PC)inDanshenpolyphenolicacidinterventiongroupwassignificantlyhigherthanthatinmodelgroup.Therefore,Danshenpolyphenolicacidcanimproveglucosemetabolismdisorderinratswithhepaticfibrosisthroughinfluencinggeneexpression,andhaspotentialtherapeuticandpreventiveeffectsonhepaticfibrosis.
