大鼠骨髓间充质干细胞向神经元样细胞诱导分化的研究的开题报告
【摘要】
骨髓间充质干细胞(BM-MSCs)是一种广泛存在于多种组织中的成体干细胞,具有广泛的多向分化潜能。BM-MSCs 在各种疾病治疗中具有重要的作用,尤其在神经系统疾病治疗中发挥了独特的作用。然而目前并没有一种有效的诱导方式将BM-MSCs向神经元分化。因此,本研究旨在通过寻找合适的培养条件,诱导BM-MSCs向神经元样细胞分化。
本研究采用大鼠BM-MSCs作为实验对象,通过不同的化学物质和因素(如转染Nurr1,添加抑制神经元分化因子的生长因子,以及辅助细胞的添加等)进行诱导分化。同时,通过荧光显微镜、RT-PCR、Western Blot等实验技术检测BM-MSCs向神经元样细胞分化的程度。
初步实验结果显示,在添加相应培养条件下,顺利地对BM-MSCs进行诱导分化,并且成功地实现了BM-MSCs向神经元样细胞分化。而且,诱导分化的BM-MSCs表达了一系列与神经元有关的标记基因,如神经元特异性标志物MAP2和Nestin,以及GABA能神经元标志物GAD67。
综上所述,本研究意在探索适宜的培养条件和诱导方法,以期进一步完善BM-MSCs向神经元分化的方式,从而为神经系统疾病治疗提供一种有效的细胞治疗策略。

【Abstract】
Bone marrow mesenchymal stem cells (BM-MSCs) are a type of adult stem cells widely present in various tissues, which have broad multi-directional differentiation potential. BM-MSCs have been shown to play an important role in treating various diseases, especially in the treatment of neurological diseases. However, there is still no effective way to induce BM-MSCs to differentiate into neurons. Therefore, this study aims to induce BM-MSCs to differentiate into neuron-like cells by finding suitable cultivation conditions.
In this study, rat BM-MSCs were used as the experimental object, and induction differentiation was performed through different chemical substances and factors (such as transfection of Nurr1, addition of growth factors that inhibit neurogenesis, and addition of assistant cells). At the same time, the degree of BM-MSCs differentiation into neuron-like cells was detected by fluorescence microscopy, RT-PCR, Western Blot and other experimental techniques.
Preliminary experimental results showed that under the appropriate culture conditions, BM-MSCs were successfully induced to differentiate into neuron-like cells. And, the induced differentiated BM-MSCs expressed a series of neuron-related marker genes, such as MAP2, Nestin, and GAD67.
