生物技术通报 ·研究报告· BIOTECHNOLOGY BULLETIN 年第 期 2014 2 沙根骨中碳酸钙矿化微生物分离筛选及鉴定 胡伟莲  戴德慧 (浙江科技学院 生物与化学工程学院,杭州 310023) 摘 要 : 沙根骨 (自命名)是在宁夏中卫地区发现的以碳酸钙为主的胶结物质将周围的沙子凝聚在一起形管状物。对沙根 骨中碳酸钙矿化微生物进行了分离筛选,共分离到12 株菌株,其中3# 菌株有较强碳酸钙矿化能力,在经1.0% CaCl2 液体培养基 中振荡培养的72 h 后,能形成矿物沉淀并形成薄膜挂壁。沉淀物质经扫描电镜 (SEM )及X 射线粉末衍射 (XRD )分析,确定为 方解石和球霞石混合晶型的碳酸钙晶体。根据其形态特征、生理生化以及 16S rDNA 对3# 菌株进行了鉴定,初步认定了该菌为类 芽孢杆菌属,命名为Paenibacillus sp.s3 ,为进一步研究沙根骨的形成机理奠定了良好基础。 关键词 : 沙根骨 生物矿化 微生物 类芽孢杆菌属 , The Isolation Screening and Identification of CaCO Mineralization 3 Strain from Sand Boon Hu Weilian Dai Dehui (Dep artment of Biology and Chemical Engineering ,Zhej iang Universtity of Science and Technology ,Hangzhou 310023 ) Abstract:  Sand bone (named by the research team )is a tube that is agglutinated by sand with cementing material mainly composed of calcium carbonate. It was found in Zhongwei area of Ningxia Autonomous Region. Strain 3# has strong ability of CaCO3 mineralization among 12 strains isolated from sand bone. Mineral deposit was produced and covered the flask wall after 72 h shake cultivation with 1.0% CaCl liquid 2 medium. To analyze the mineral deposits, XRD and SEM were performed. The result was showed that mineral deposit was made up of calcium carbonate, which contains two crystal forms (calcite and vaterite ). Based
