硕士学位论文 硕士学位论文 摘要 第四部分主要研究APFM对四氧嘧啶所致糖尿病小鼠的降糖降脂作用:实验 结果显示,随着给药时间的延长,APFM能显著减轻糖尿病小鼠多饮、多食、多 尿的症状,降低糖尿病小鼠的血糖值。且中剂量组的效果最佳,四周后其血糖值 与正常组比无显著差异,血糖曲线下面积与模型组有显著差异(P0.05)。APFM 还能减少小鼠血清中的NO、NOS、TL、TC含量,提高血清中HDL含量。表明 APFM能有效降低糖尿病小鼠的高血糖和高血脂。 关键词:桑叶:酸性蛋白多糖;分离纯化;理化性质;生物学活性。 II 硕士学位论文 硕士学位论文 Abstract Study on Separation,Purification,Characterization and Biological activities of acid proteoglycan from Folium mori Abstract:In this paper,we separated and purified an acid proteoglycan from Folium mori(APFM),also,the Characterization and Biological activities of this acid proteoglycan be researched. The experiment included four parts: In the first part,we studied the extraction,separation,purification and characterization of APFM:The polysaccharides was extracted from Folium mori by optimizing water,then isolated and purified by D101resin,DEAE—cellulose column and SephadexG一100 gel column chromatography.IR,UV,HPLC,13C-NMR,1H-NMR and Amino acids composition identification were used for structural analysis.The results showed that APFM WaS a pure acid proteoglycan,composed of uron/c acids, D.Rhamnose、L.Arabinose、D—Fructose、D.Glucose and D-Galactose(the molar ratio:8.91:2.71:1.00:3.75:6.04).It’S monomer of component was Q—pyranose.In addition,APFM contained seventeen kinds of amino acids,seven kinds of which were human essential amino acids. In the second part,the antioxidant activities of APFM was studied:Results showed that APFM could efficiently scavenge HO·and‘02。produced in chemical mimicking system,inhibit the production and accumulation of MDA in mice organs induced by HO·and in the organs of diabetic mice induced by alloxan,prominently lower the swelling of mice liver mitochondria induced by HO‘,also increase SOD activity in dose.dependent manner.Conclusion:APFM had strong antioxidant activity in vitro and in vivo,it WaS an efficient fre
