葡萄籽原花青素对人卵巢癌裸鼠移植瘤细胞增殖和凋亡的影响 徐 蕾,钟 玲,胡琴琴,蒋兴伟,池余刚(重庆医科大学附属第二医院妇产科,重庆 400010) 摘要: 目的 探讨葡萄籽原花青素对人卵巢癌裸鼠皮下移植瘤增殖和凋亡的影响和机制。方法 建立24只人卵巢癌裸鼠皮下移植瘤模型,随机分成3组,每组8只:(1)原花青素高浓度组(200 mg/kg);(2)原花青素低浓度组(100 mg/kg);(3)空白对照组。观察各组裸鼠皮下移植瘤生长情况,HE染色观察肿瘤细胞形态,流式细胞分析细胞凋亡,RT-PCR,Westrnblot检测肿瘤细胞Bax,Caspase-3在RNA及蛋白水平的表达情况。结果 原花青素组皮下移植瘤抑瘤率分别为34.303、28.594,与对照组相比差异具有显著性(P0.05);治疗组肿瘤细胞出现明显的凋亡形态学变化,流式细胞检测治疗组肿瘤细胞凋亡率分别为67.965,24.650;原花青素还可以明显增高Bax,Caspase-3 RNA和蛋白的表达,且呈浓度依赖性。结论 葡萄籽原花青素对卵巢癌皮下移植瘤生长有抑制作用,并促进其凋亡。其作用可能通过增强Bax,Caspase-3的表达而实现。 ;R737.31 文献标识码:A Grape seed proanthocyanidin inhibits human ovarian cancer xenograft in nude mice XU Lei, ZHONG Ling, HU Qin-qin, JIANG Xing-wei ,CHI Yu-gang (Department of Gynaecology and Obstertrics, Second Affliated Hospital, Chongqing Medical University,Chongqing 400010, China ) Abstract: Objective To study the effects of grape seed proanthocyanidin extract (GSP) on the proliferation and apoptosis of the transplanted tumor of human ovary cancer in nude mice and to explore the implying molecular mechanism. Methods A human ovarian cancer model was established by subcutaneously human ovary cancer cell line SKOV3 into nude mice. The 24 mice were randomly and equally divided into 3 groups, the high-dose GSP group (200 mg/kg, HD GSP), the low-dose GSP group (100 mg/kg, LD GSP) and the control group (Normal saline). GSP solution or normal saline was intragastically given to the mice at 10 mg/kg. Tumor volume and weight were measured. HE staining was used to observe the tumor cells morphology. Apoptosis of the tumor cells was analyzed by flow cytometry (FCM), semi-quantitative RT-PCR was used to detect the mRNA expressions of Caspase-3 and Bax, and their protein expressions detected by Western blot analysis. Results The inhibitory rate in the HD and LD GSP groups were 34.303% and 28.594% respectively, which was significantly higher than that in the control group (P0.01). The apoptotic rate in the GSP groups were 67.965% and 24.650% respectively,
