莪术醇对小鼠的急性毒性研究 【摘要】 目的:观察高纯度莪术醇对小鼠所产生的反应、死亡情况等急性毒性作用,测定其在最大允许浓度和体积下的最大耐受剂量(MTD),为拟订临床使用安全剂量提供实验依据。方法:将40只健康小鼠随机分为对照组和试验组进行急性毒性试验,其中试验组以最大允许浓度和体积的给药量(100.00mg/ml,0.4ml/20g )给药,连续观察7天。结果:按照2.00g/(kg·次)给药,连续2次,所得MTD大于4.00g/kg(相当于成人439.8倍剂量),给药后7天小鼠精神状态良好、毛色光滑、饮食、粪便正常,未出现任何毒性反应和小鼠死亡,鼠体质量增长比较无显著性差异,第8天处死动物观察内脏各器官无明显改变,主要脏器指数无显著性差异。结论:高纯度莪术醇的最大耐受剂量MTD大于4g/kg (相当于成人439.8倍剂量),表明莪术醇是低毒性的药用活性物质,其临床拟用剂量是安全的。 【关键词】The effects of alcohol in mice with acute toxicity research Year 2009’s Pharmacy Student ChenXin Supervisor ChenXU [Abstract] Objective: to observe the high purity curcumenol produced by the reaction in mice, acute toxic effects such as death, measure its under the maximum allowable concentration and volume of the maximum tolerated dose (MTD) and provide experimental basis for clinical use safe dose. Results: according to the 2.00 g/(kg ·time), 2 times in a row, the MTD is greater than 4.00 g/kg (equivalent to of adult dose), 7 days after the treatment in mice smooth good mental state, color, food, faeces is normal, without any toxic effects and mice died, mice increase in body mass is no significant difference, day 8 kill animals to observe no significant changes in the visceral organs mainly viscera index there was nosignificant difference. Conclusion: high purity curcumenol maximum tolerance dose of MTD is greater than 4 g/kg (equivalent to of adult dose), showing curcumenol and clinical decodes dose is safe.
