摘要 糖化酶是工业生产中应用最为广泛的酶制剂之一。目前,糖化酶已广泛应用于食品等许多行业。本设计本设计通过对年产2000吨的黑曲霉生产糖化酶的工艺流程进行设计,综述了糖化酶和米曲霉的应用和发展,比较了盐析法和超滤法提取糖化酶的优缺点。以原有的生产工艺为基础,本设计中糖化酶生产采用玉米粉作为碳源,豆饼为氮源,以米曲霉作为生产菌种,运用深层液体发酵法。设计中重点改进的部分在下游提取工艺,它以超滤浓缩的方法替代以前盐析提取糖化酶的工艺。 此次设计以年产2000吨糖化酶为生产规模,同时论文中做出了生产工艺流程图,进行了物料衡算和设备选型,设计了发酵罐和种子罐的尺寸,分析比较了盐析和超滤两种提取工艺的经济效益,得出了超滤工艺提取糖化酶优于盐析法提取工艺。 关键词:糖化酶,米曲霉,生产工艺,盐析,超滤 Abstract Glucoamylase is one of the most widely used in industrial production of enzymes.At present, the glucoamylase has been widely used in many industries such as food industrial. The Design review application and development of the glucoamylase and Aspergillus oryzae, and compared the advantages and disadvantages of salting method and ultrafiltration extraction of glucoamylase.On the base of the original production process, the design of glucoamylase production adopts corn flour as the carbon sources,soybean meal as nitrogen source, Aspergillus oryzae as a production strain, and use the deep liquid fermentation.Design focus on improving part in downstream extraction process, which concentrated by ultrafiltration method to replace the previous salting-out extraction of glucoamylase process. The scale of production of the design is 2000 tons of glucoamylase, at the same time the paper made a production process flow diagram,the material balance and the selection of equipment, and the size of fermentation tank and seed tank are designed, economic benefit of two extraction methods adopting the salting out and ultrafiltration is analysised and compared, we can see the UF process for extraction of glucoamylase is better than salting out extraction process.
