Introduction:
Hemolyticphosphatidylcholine(HPC)isaphospholipidmoleculethatisgeneratedfromthebreakdownofredbloodcellmembranes.HPChasbeenshowntohaveasignificanteffectontheproliferationofvascularsmoothmusclecells(VSMCs).However,thesignalingpathwaysinvolvedinthisprocessarenotwellunderstood.Inthisstudy,weaimtoinvestigatetheproliferativeeffectsofHPConVSMCsandtheassociatedsignalingpathways.
Objectives:
1.ToexaminetheeffectofHPConVSMCproliferation.
2.ToexplorethesignalingpathwaysthatmediatetheproliferativeeffectofHPC.
3.TodeterminethespecificreceptorsinvolvedinHPC-inducedVSMCproliferation.
Methods:
CulturedVSMCswillbetreatedwithHPCatvariousconcentrations.ThecellproliferationratewillbemeasuredusingtheMTTassay.Westernblotwillbeusedtodetecttheexpressionofproliferativesignalingmolecules,includingERK,AKT,andSTAT3.RT-PCRwillbecarriedouttoanalyzemRNAexpressionofspecificreceptors.
ExpectedResults:
WeexpecttofindthatHPCenhancesVSMCproliferationandthatthiseffectismediatedbytheactivationoftheERK,AKT,andSTAT3signalingpathways.Additionally,weanticipateidentifyingthespecificreceptorsinvolvedinHPC-inducedproliferation.
Conclusion:
ThefindingsofthisstudymayhelpelucidatethemechanismbywhichHPCpromotesVSMCproliferation,providinginsightsintopotentialtherapeutictargetsforvasculardiseases.
