广西圆斑蝰蛇毒凝血因子X激活剂的分离纯化及其止血作用的研究的开题报告

【摘要】

广西圆斑蝰是中国特有的毒蛇之一。圆斑蝰蛇毒在血凝系统中产生广泛作用，能够促进血小板的活化和血凝时因子X的活化。本研究旨在通过分离纯化圆斑蝰蛇毒中的止血因子X激活剂，研究其在体内的止血作用及相应机制，为后续开发抗血栓药物提供理论支持。

该研究将通过以下步骤实现：

（1）从广西圆斑蝰蛇毒中利用吸附剂进行分离纯化。

（2）将纯化后得到的圆斑蝰蛇毒毒液用于体内实验，检测其对小鼠出血时间的影响。

（3）运用Westernblot等方法，检测止血因子X激活剂与小鼠血浆中凝血因子X的相互作用。

（4）通过体内实验，探寻止血因子X激活剂在体内促进血小板活化和血凝时因子X的活化的作用。

【Abstract】

TheTrimeresurusstejnegeri,avenomoussnakeindigenoustoChina,hasawiderangeofeffectsonthebloodcoagulationsystem,specificallyactivatingfactorXduringplateletactivationandclotting.ThisstudyaimstoisolateandpurifytheXactivatorofthesnakevenomofTrimeresurusstejnegeri,andtoinvestigateitshemostaticeffectandcorrespondingmechanisminvivo,providingtheoreticalsupportforthedevelopmentofantithromboticdrugs.

Thestudywillbeconductedinthefollowingsteps:

(1)TheTrimeresurusstejnegerivenomwillbepurifiedusinganadsorbent.

(2)Thepurifiedvenomwillbeusedforinvivoexperimentstodetermineitseffectonthebleedingtimeofmice.

(3)TheinteractionbetweentheXactivatorandfactorXinmouseplasmawillbestudiedusingWesternblotandothermethods.

(4)TheeffectofXactivatoronplateletactivationandfactorXactivationduringclottingwillbeinvestigatedusinginvivoexperiments.
