杨树抗逆相关CDPK基因家族分析及PtELP1基因功能研究的中期报告
摘要:
本研究旨在探索杨树抗逆相关蛋白激酶(CDPK)基因家族的进化和功能,并研究一个新的抗逆基因PtELP1的功能。通过对杨树基因组数据库的检索和生物信息学分析,本研究鉴定了17个CDPK基因家族成员和一个PtELP1基因,并对它们的结构、进化关系、保守区域、基因表达及功能进行了初步分析。
结果表明:研究杨树中的CDPK基因家族在结构和保守区域上存在较大差异,并在演化上分化为四个亚家族。其中,A亚家族基因含有一个保守的N端CA结构域,而BCD亚家族基因则包含一个保守的C末端结构域。所有CDPK基因家族成员均在杨树不同的组织中表达,并响应多种逆境胁迫,如盐、干旱、寒冷和病毒感染等。此外,我们还鉴定了一个新的PtELP1基因,它可能在杨树抗逆过程中发挥重要作用。进一步研究发现,PtELP1基因的过表达可显著增强杨树在盐胁迫下的生长和幸存能力,并促进根系的生长和发育。
综上所述,本研究为深入理解杨树CDPK基因家族的进化和功能及PtELP1基因在杨树抗逆过程中的作用提供了初步证据和工具。

Abstract:
The aim of this study was to investigate the evolution and function of calcium-dependent protein kinase (CDPK) gene family related to stress resistance in poplar, and to study the function of a new stress-resistant gene PtELP1. Through searching and bioinformatics analysis of the poplar genome database, we identified 17 CDPK gene family members and one PtELP1 gene, and conducted preliminary analysis of their structure, evolutionary relationship, conserved region, gene expression and function.
The results showed that there were considerable differences in the structure and conserved regions of the CDPK gene family in poplar, and they were divided into four subfamilies in evolution. The A subfamily genes contained a conserved N-terminal CA domain, while the BCD subfamily genes contained a conserved C-terminal domain. All CDPK gene family members were expressed in different tissues of poplar, and responded to various stress conditions, such as salt, drought, cold and viral infection, etc. In addition, we identified a new PtELP1 gene, which may play an important role in poplar stress resistance. Further studies revealed that overexpression of the PtELP1 gene significantly enhanced the growth and survival ability of poplar under salt stress, and promoted the growth and development of roots.
