存活素血管内皮生长因子C在结肠癌中表达及其临床意义 　　【摘要】 目的 探讨结肠癌组织中存活素（survivin）、血管内皮生长因子-C（VEGF -C）的表达，及其相互关系和临床意义。方法 应用免疫组化法检测35例结肠癌组织和15例癌旁组织中survivin及VEGF-C的表达水平。结果 结肠癌组织中survivin、VEGF-C阳性表达率分别为60．0%、65．7%显著高于癌旁非癌组织的13．3%（P0．05）。survivin、VEGF-C的表达呈正相关（P0．01）。结论 结肠癌组织中survivin、VEGF-C表达均增高并且呈正相关。两者在结肠癌淋巴结转移中有协同效应，共同促进结肠癌的淋巴转移。? 　　【关键词】结肠癌;存活素;血管内皮生长因子-C;免疫组化 　　 　　Expression and Clinical Significance of survivin，VEGF -C in Human Colon Cancer 　　 　　WANG Xiao-ling,HE Jian-qin,LI Jun-da.Department of Gastroenterology,The Forth Peoples Hospital of Shenzhen,Shenzhen 518033,China 　　? 　　【Abstract】 Objective To investigate the relationship between the expression of survivin,VEGF-C and the clinicopathologic parameters in colon cancer.And evaluate the clinical significance.Methods 35 colon cancer tissues,15 adjacent noncancerous specimens were obtained.Survivin,VEGF-C protein expression were evaluated by immunohistochemical analysis.Results Survivin and VEGF-C were overexpressed in colon camcer tissues (60．0%,(65．7%) were significantly higher than those in noncancerous tissues(13．3%)(P0．05).There was positive correlation between the expression of survivin and VEGF-C(P0．01).Conclusion Survivin overexpression was significantly associated with elevated VEGF-C.The co-expression of survivin and VEGF-C might play an important role in lymphatic spread of colon cancer.? 　　? 　　【Key words】Colon cancer survivin Vascular endothelial growth factor-C (VEGF-C) immunohistochemistry 　　 　　存活素(survivin)是新近发现的一种凋亡抑制基因,在多种恶性肿瘤组织中异常高表达,可增强肿瘤细胞的转移潜能。血管内皮生长因子-C(vascular endothelial growth factor-C,VEGF-C)是VEGF家族的新成员，又称为淋巴管生长因子，诱导淋巴上皮增殖和淋巴管增生，在肿瘤的淋巴管播散中起重要作用。本研究应用免疫组化技术，检测结肠癌组织中survivin、VEGF-C的表达，探讨其与结肠癌生物学行为的关系及临床意义。 　　 　　1 材料和方法 　　 　　1．1 标本与试剂 收集结肠癌标本35例，癌旁非癌组织15例（距癌灶边缘大于5cm），均未接受放化疗。其中男性25例，女性10例。年龄29岁~72岁，中位年龄52．7岁。标本常规福尔马林固定，石蜡包埋，3~4 μm连续切片分别进行HE染色和免疫组织化学研究。根据1987年国际抗癌联盟公布的PTNM分期标准进行TNM分期。? 　　兔抗人survivin多克隆抗体（Santa Cruz,U.S.A）工作浓度为1∶50，兔抗人VEGF-C多克隆抗体（Zymed,U.S.A）工作浓度为1∶80。SP试剂盒购于北京中山生物技术有限公司。 　　1．2
