5-氟尿嘧啶衍生物的合成与结构确证
摘 要:目的:合成5-氟尿嘧啶的衍生物以及其结构的确证。方法:1. 以5-氟尿嘧啶,六甲基硅胺烷,三甲基氯硅烷为原料反应得到二硅醚化5-氟尿嘧啶,再加入二溴乙烷,得到N1-2-溴乙烷取代5-氟尿嘧啶。2.以5-氟尿嘧啶为原料,在一定的条件下加入溴乙酸,碱性条件下反应得到5-氟尿嘧啶-1-基乙酸。3. 以5-氟尿嘧啶与甲醛为原料,在一定条件下得到中间产物1, 3-二羟甲基-5-氟尿嘧啶,继而与肉桂酸酯化,最终得到肉桂酸5-氟尿嘧啶甲酯。结果与结论:通过对5-氟尿嘧啶结构修饰,合成了三种目标产物,其结构经过TLC、IR、MS等得到确证。

The synthetic and Structure confirmation of 5-fluorouracil derivatives
Abstract: Objective: 5-Fluorouracial is a kind of effective anticancer agent used in clinic and to control tumor cells obviously. But it also has stronglytoxic side-effect. In order to obtain lowly toxic anticancer agents, 5-FU derivatives were prepared. Methods: Three types of FU derivatives were synthesized. Methods: 5-Fluorouracil-1-yl acetic acid was synthesized from bromoacetic acid and 5-fluorouracil in 83.5% yield; cinnamylate 5- fluorouracil-1yl-methyl ester were synthesized by two steps with 5-fluorouracil and cinnamylate as the raw materials. Results: The structures were confirmed by IR, TLC, MS. Conclusions 5-fluorouracil-yl-acetic acid, N1-(2- bromoethyl) -fluorouracil, and cinnamylate 5-fluorouracil-1yl-methyl ester are promising derivatives of 5-fl
