Introduction:
Breastcanceristhemostcommoncancerdiagnosedinwomenandaleadingcauseofcancer-relateddeathworldwide.Despiteadvancesintherapy,themortalityrateassociatedwiththisdiseaseremainshigh.Therefore,noveltherapeuticapproachesthatcanimprovesurvivalandqualityoflifeforpatientsareurgentlyneeded.Naturalcompoundsderivedfromplantshavegainedincreasingattentionaspotentialanticanceragentsduetotheirlowtoxicityandhighefficacy.D-pinitol-β-ivyglycoside(D-PIG)isolatedfromHederahelixisanaturalcompoundwithpotentanticancerproperties,includingtheinductionofapoptosisinbreastcancercells.However,theprecisemechanismsofD-PIG-inducedapoptosisinbreastcancercellsarenotfullyunderstood.TheobjectiveofthisstudyistoinvestigatethemolecularmechanismsunderlyingD-PIG-inducedapoptosisinbreastcancercells.
Methods:
ThehumanbreastcancercelllineMCF-7willbeusedinvitrotoinvestigatetheeffectsofD-PIGoncellviability,apoptosis,andrelatedmolecularmechanisms.Thefollowingexperimentalmethodswillbeperformed:
1.Cellviabilityassay:MCF-7cellswillbetreatedwithvariousconcentrationsofD-PIGfor24or48hours,andcellviabilitywillbemeasuredbyMTTassay.
2.Apoptosisassay:MCF-7cellswillbetreatedwithD-PIGfor24or48hours,andapoptosiswillbeevaluatedbyAnnexinV/PIstainingfollowedbyflowcytometryanalysis.
3.Westernblotanalysis:Theexpressionlevelsofvariousapoptosis-relatedproteins,suchascaspase-3,caspase-9,PARP,andBcl-2,willbeexaminedbywesternblotanalysis.
4.Real-timePCR:ThemRNAlevelsofvariousapoptosis-relatedgenes,suchascaspase-3,caspase-9,andBcl-2,willbequantifiedbyreal-timePCR.
Expectedresults:
WeexpectthatD-PIGwillinduceapoptosisinMCF-7cellsinadose-dependent
