摘 要:利用离子交换层析和凝胶层析对浒苔多糖进行分离纯化,并探讨其磷酸化修饰的最佳工艺条件。通过单因
素试验确定反应温度、反应时间、反应p H 值、磷酸化试剂质量浓度的取值范围,再用响应面设计法确定磷酸化修
饰的最佳条件。结果表明:反应时间、磷酸化试剂质量浓度显著影响浒苔多糖磷酸化程度,当反应温度8 0 ℃、反
应时间5h 、反应pH9.0 、磷酸化试剂质量浓度0.10g/mL 时,磷酸化修饰的浒苔多糖磷酸根含量( 以P 计)达到14 .40% 。

Abstract :This study was undertaken to optimize the phosphorylation of Enteromorp h polysaccharides prepared by hot water
extraction, deproteinization , decolorization, and sequential chromatographic purification on DEA-Sepharose Fast Flow and
Sephadex G- 100 columns. Phosphate group content in phosphorylated products was investigated with respect to reaction
condition s including temperature, reaction time, pH and phosphorylation agent dosage (sodium tripolyphosphate-sodium
trimetaphosphate at a mass ratio of 6 :1) by one-factor-at-a-time experiments and subsequent respon se surface analy sis. The
optimal phosphorylation conditions were pH 9.0, temperature 80 ℃and phosphorylation agent dosage 0.10 g/mL for a reaction
