Introduction
Theaccumulationofamyloid-β(Aβ)peptidehasbeenconsideredacriticalcontributortothepathogenesisofAlzheimersdisease(AD),aneurodegenerativedisordercharacterizedbyprogressivecognitivedeclineandneuroinflammation.Aβinjuryleadstoneuroinflammationandimpairsthemechanismsofbrainneurosteroidsynthesisandmetabolism,whichhaveanimpactonvariousneurologicalfunctionsandbehavior.Therefore,studyingtheimpactofAβinjuryonneurosteroidmetabolismandunderstandingtheprotectiveroleofprogesteroneagainstAβ-inducedneuroinflammationarecriticalstepstowarddevelopingeffectivetherapeuticapproachesforAD.
Objectives
TheprimaryobjectiveofthisresearchistoinvestigatethechangesinneurosteroidsynthesisandmetabolismintheratbrainfollowingAβinjuryandtodeterminethemechanismthatunderliesthesechanges.ThestudyalsoaimstoexaminetheneuroprotectiveeffectsofprogesteroneonAβ-inducedneuroinflammationanditsrelationshipwithneurosteroidmetabolism.
Methodology
ThisstudywillinvolvetheuseofmaleWistarrats,whichwillbedividedintofourgroups(A,B,C,andD).GroupAwillserveasthecontrolgroupandwillreceiveasalineinjection.GroupBwillreceiveasingleinjectionofAβ,whichwillinduceneuroinflammation.GroupCwillreceivechronicprogesteronetreatment(20mg/kg/day)fortwoweeksbeforetheAβinjectiontodeterminetheprotectiveeffectsofprogesteroneonthebrainagainstAβinjury.GroupDwillreceiveacombinationofAβandprogesteronetreatment.
Thelevelsofneurosteroids,suchaspregnenolone,progesterone,andallopregnanolone,willbemeasuredinthebraintissueusingliquidchromatographyandtandemmassspectrometry.Theexpressionlevelsofgenes
