The results show that this technique can significantly enhance the arctigenin content in fructus arctii. The optimum medium components are 015 g fructus arctii powder, 3 g bran, 2 g bagasse, 013 g peptone, and 10 ml Mandels nutr-i ent fluid. Solid-to-liquid ratio is 1B316 g/ml. Initial pH of the medium is 516. Lower yield was observed with higher initial arctiin concentration used. The yield reached 9310% when 01174 mmol/L arctiin was hydrolyzed at 30efor 7 d. In 7 d, the hydrolysis of arctiin is in accordance with the equation of Monod. The kinetic constants areVm=01083 7 mmol/(L#d) andKm=0116 mmol/L. Key words: arctiin; arctigenin;B-glucosidase; Chinese medicine; fermentation;Aspergillus nigerZJUT712 发酵中药是现代生物技术和中药研究的结合, 为中药的发展开辟了新空间[1].牛蒡子是菊科植物 牛蒡(Arctium LappaL.)的干燥成熟果实.牛蒡子 苷及牛蒡子苷元是牛蒡子发挥药效的重要成分,具 有利尿、泻下、降血压、抗癌抗突变、抗老年性痴呆和 治疗肾病综合症等作用[2~17].牛蒡子的药效与牛 蒡子苷在体内的代谢过程具有相关性,牛蒡子苷在 消化道中被肠道菌转变为苷元的脱甲基化合物,此 脱甲基化合物在肝脏儿茶酚-O-甲基转移酶的作用 下恢复为苷元,苷元被血液输送到各个器官发挥作 用[18].由于牛蒡子苷元可直接被人体吸收而发挥 药效,因此在体外将牛蒡子苷转化为牛蒡子苷元将 有助于促进牛蒡子摄入人体并快速起效.
