一、研究背景
雌激素受体playsanessentialroleinfemalereproduction,development,andphysiology.Estrogenreceptorα(Erα)isthemostpredominantisoformoftheestrogenreceptorandisakeyregulatorofestrogen-mediatedtranscriptionalsignaling.DysregulationofErα-mediatedtranscriptioncanleadtovariouspathologicalconditions,suchasbreastandendometrialcancer.Therefore,understandingtheregulatorymechanismsthatcontrolErα-mediatedtranscriptioniscrucialforthedevelopmentoftherapeuticstrategiesagainstestrogen-relateddiseases.
KIAA1915isanuclearproteinthathasbeenimplicatedintheregulationoftranscriptionalactivity.RecentstudieshavesuggestedapotentialroleforKIAA1915inmodulatingestrogensignaling.However,thespecificmechanismsbywhichKIAA1915regulatesErα-mediatedtranscriptionanditsfunctionalconsequencesarestillunclear.
二、研究目的
TheaimofthisstudyistoinvestigatetheroleofKIAA1915inregulatingErα-mediatedtranscriptionandelucidatethemolecularmechanismsinvolved.Specifically,wewill:
1.ExaminetheeffectsofKIAA1915onErα-mediatedtranscriptionalactivityusingaluciferasereporterassay.
2.IdentifytheKIAA1915-interactingpartner(s)involvedinregulatingErα-mediatedtranscriptionusingco-immunoprecipitationandmassspectrometryanalysis.
3.DeterminetheimpactofKIAA1915-mediatedregulationofErα-mediatedtranscriptiononestrogensignaling-relatedoutcomes,suchascellproliferationandmigration.
三、研究方法
Toachieveourresearchobjectives,wewillusethefollowingmethods:
1.Constructionofexpressionvectors:WewillsynthesizeandconstructvectorsexpressingKIAA1915andErα.
2.Luciferasereporterassay:WewilltransfectdifferentcombinationsofvectorsintoMCF-7cells,ahumanbreastcancercelllineexpressingendogenousErα,andusealuciferasereporterassaytoexaminetheeffectsofKIAA1915onErα-mediatedtranscriptionalactivity
