Introduction:
Alzheimersdisease(AD)isachronic,neurodegenerativedisorderthataffectsmillionsofpeopleworldwide.OneofthepathologicalfeaturesofADistheaccumulationofamyloid-β(Aβ)peptidesinbraintissue.RecentstudiessuggestthatvaccinationagainstAβmaybeapotentialtherapyforAD.However,previousclinicaltrialsusingfull-lengthAβastheantigenhavenotshownsatisfactorytherapeuticbenefits.Toaddressthisissue,weproposetodevelopamulti-epitoperecombinantAβvaccinethattargetsmultipleAβepitopestoelicitamoreeffectiveimmuneresponse.
Objectives:
1.TocloneandexpressmultipleepitopesofAβasrecombinantfusionproteins
2.Topurifyandassembletherecombinantfusionproteinsintoamulti-epitopevaccine
3.Toevaluatetheimmunogenicityofthemulti-epitopevaccineinvitroandinvivo
Methods:
1.BioinformaticsanalysisofAβepitopestoselectcandidateepitopesforthevaccine
2.Codinggenesfortheselectedepitopeswillbesynthesizedandclonedintoexpressionvectors
3.Expressionandpurificationoftheepitope-fusionrecombinantproteins
4.Purificationandassemblyoftherecombinantproteinsintoamultimericvaccine
5.Immunizationofmicewiththemulti-epitopevaccine,measuringtheirantibodyresponseandTcellresponse
Expectedoutcomes:
Weanticipatethatthemulti-epitopeAβvaccinewillelicitastrongerimmuneresponsecomparedtofull-lengthAβvaccination.OurstudymayofferanewapproachfordevelopinganeffectivetherapeuticstrategyforAD.
Conclusion:
Theestablishmentofamulti-epitoperecombinantAβvaccineandtheevaluationofitsimmunogenicitywillprovideinsightintothedevelopmentofamoreefficaciousvaccineforAD.
