紫外可见分光光度计
紫外可见分光光度计原理是分子的紫外可见吸收光谱是由于分子中的某些基团吸收了紫外可见辐射光后,发生了电子能级跃迁而产生的吸收光谱。它是带状光谱,反映了分子中某些基团的信息。可以用标准光谱图再结合其它手段进行定性分析。根据Lambert-Beer定律:A=εbc,(A为吸光度,ε为摩尔吸光系数为液池厚度,c为溶液浓度)可以对溶液进行定量分析。你可以用紫外可见分光光度计测定定三种农药的波长在某溶液中的最大、最小吸收波长。配制溶液-在光谱检测项下进行-调整检测光谱范围及速度--扫描光谱图--吸光度最大处对应波长为最大吸收波长,吸光度最小处对应的波长为最小吸收波长。
测试过程
① 数字显示透光度100.0(或吸光度0.00)2~3s后,将盛有标准溶液的比色皿移至光路,打印系统便自动打印出所得数据;
② 将盛有样品溶液的比色皿移至光路,打印系统即自动打印出该样品的数据.待第一个样品数据打印完毕后,将第二个样品置于试样室光路………,若有多个样品,操作以此类推。
